The HUMANKIND Cultures of Latin America Pack was released on June 9, 2022 introducing a variety of new content inspired by Latin American history and culture. It includes six new playable cultures, each representing different historical eras.

These include the Caralans (Era 1), an ancient builder civilization known for urban and agricultural advancements; the Nazca (Era 2), renowned for their ceramics and geoglyphs; the Taíno (Era 3), agrarians who thrived in the Greater Antilles; the Inca (Era 4), expansionists who established the Tawantinsuyu empire; the Argentinians (Era 5), merchants who became global exporters of agricultural goods; and the Cubans (Era 6), aesthetes who gained prominence after the 1959 revolution.

In addition to these cultures, the DLC adds six new wonders including the Pyramid of the Sun in Teotihuacan, Maracanã Stadium, Salar de Uyuni, Salto Ángel, Lençóis Maranhenses, and the Atacama Desert. Players also gain access to nine independent peoples, 15 narrative events, and over 30 new themed music tracks composed by Arnaud Roy, further enriching the gameplay experience.
